N2 - In this chapter we present a MATLAB-based computational pipeline for the quantification of monolayer migration assays. Wound healing assay (or scratch assay) is a commonly used in vitro assay to assess collective cell migration. Our pipeline outputs traditional and spatiotemporal readouts that quantify the group migration properties and was previously used for a screen that included thousands of time-lapse sequences. You will learn how to execute the pipeline, the principles behind the design and implementation choices we made, pitfalls, tips, and tricks in using it.
